It's just a funny word said in a funny way when you try to use the bottle on yourself. As if it's the sound effect. Nothing more. Obviously entertained Paw quite a bit.

You never played the KQ2 AGD remake? You owe it to yourself as a King's Quest fan to play it. I love how they gave sense to the whole "Black Cloak Plot" from KQ6 that never got really explained.

That's what most people dislike about it. To them I keep saying to just look at it as a well-done and well-executed alternate experience. It's not canon and never will be. It's just a fun game that has a lot more fun puzzle gameplay and interesting writing than the original has. It really plays like a classic Sierra game in pretty much every way.

The whole joke is there in the video. There's no further exposition beyond "click bottle on self." If the sound of an uninterested narrator blatantly saying "donk" doesn't amuse you, then I don't know what to say.

I think you may not know what to say then... I believe the humor is lost on me, in this case.
